Về cơ chế tác dụng, riboflavin được biến đổi thành thành 2 co-enzym là FMN (flavin mononucleotid) và FAD (flavin adenin dinuclrotid). Đây là các dạng co-enzym hoạt động cần cho sự hô hấp của mô. Riboflavin cũng cần cho sự hoạt hóa pyridoxin (vitamin B6), sự chuyển tryptophan thành niacin và liên quan đến sự toàn vẹn của hồng cầu.
